Most Holy Trinity Church, located on Buell Lane, midway between Toilsome Lane and Montauk Highway, was built in 1894, but it wasn't until 1907 that it became a self-serving parrish. With the largest Christian congregation in East Hampton, it serves 1,700, including a large Latino contingent. Apostolado Hispano give masses there in Spanish. Some masses are held in the Parish Hall next door that was built in 2000 because it seats 700 people, as opposed to the original church at 57 Buell Lane, which only seats about 200. The church office is next the hall, in the old convent, at 79 Buell Lane.

The former Stella Maris Regional School, which is the brick building on the either side of the church, is used for Sunday school and is rented to preschools from time to time. The Most Holy Trinity Cemetery is located 2 miles away on Cedar Street.

During the summer, the parish holds additional masses at St. Peter the Apostle in Amagansett, which was built in the 1920s.
